Methods & Procedures

Procedures and methodology used for, e.g., sampling and sample storage, sample pretreatment, extraction and analysis, including which laboratories are involved, references to methods employed, etc.

(1) tissue sampled from seals and stored/transported in liguid nitrogen (2) sample homogenised in buffer and cytosolic fractions separated (3) metallothionein analysed in cytosolic fraction by differential pulse polarography and HPLC (4) protein determined in cytosolic fraction by spectrophotometric method

QA/QC Information (what QA/QC procedures are implemented, laboratories involvment in QA/QC activities, model verification/validation routines, etc.)

NIVA is a partner in the BEQUALM programme (EU-funded) and lead country for metallothionein analyses. Mammalian metallothionein is not included in the programme at present, so internal QA procedures will be used. The protocol includes the use of reference materials.
