The influence of body lipid status on PCB toxicokinetics in the anadromous Arctic charr (Salvelinus alpinus)

The objective is to study the relationship between natural seasonal variations in body lipid status of sea migrating Arctic charr and disposition (e.g. tissue distribution)of PCB, particularly in relation to the toxical potential of a certain body burden of PCB.

The study is performed as an experimental contamination of wild, anadromous charr that are brought into the laboratory.

The proportional distribution of the total body burden of PCB between tissues containing most of the stored lipids (e.g. muscle)and vital organs such as liver, kidney and brain are quantified. Furtehrmore, the biological response to PCB in realtion to tissue distribution is studied by quantifying the activity of the hepatic P450 1A enzymes.

Methods & Procedures

Procedures and methodology used for, e.g., sampling and sample storage, sample pretreatment, extraction and analysis, including which laboratories are involved, references to methods employed, etc.

Fish are contaminated by gastric implantation. Total lipids and lipid composition are analysed by extraction and high-performance thin-layer chromatography. PCB are analysed by cyclohexane extraction and capillary gas chromatography. All analyses are performed at the NILU laboratories either at Kjeller or in Tromsø. EROD activity are quantified by standard methods (Wolkers et al., 1996. Aquat. toxicol., 35: 127-138).

PCB analyses in accordence with EU-standard EN 45001.
